MEASUREMENT OF PROGRESSIVE ANTITHROMBIN (AT III) ACTIVITY USING ANTI IIa METHOD (#221122) INTENDED USE: Chromogenic assay for the quantitative determination of the “progressive” activity of Antithrombin (AT), heparin independent, in human citrated plasma using an anti IIa

چکیده

ASSAY PRINCIPLE: Antithrombin is the major physiological coagulation inhibitor. It inhibits coagulation serine esterases, especially Thrombin, Factor Xa and Factor IXa, regulates coagulation pathway and prevents from thrombosis. In the absence of heparin, Antithrombin is a “slow acting”, progressive, inhibitor of coagulation serine esterases. The antithrombin Anti-IIa method is a method based on the inhibition of a constant amount of Thrombin (IIa), by the tested antithrombin (without addition of heparin, but with a prolonged inhibition time between Factor IIa and tested AT), and hydrolysis of a Thrombin specific chromogenic substrate, by Thrombin in excess. pNA is then released from the substrate. The amount of pNA released is then a relation of the residual Thrombin activity. There is an inverse relationship between the concentration of AT present in the tested plasma and color development, measured at 405 nm.

Novel method for measurement of heparin anticoagulant activity using SPR.

A novel method has been developed for the easy measurement of heparin's anticoagulant activity using surface plasmon resonance. The anticoagulant activity of target heparin was evaluated by measuring the competitive antithrombin III binding of analyte heparin in the solution phase and USP heparin immobilized on chip surface. Heparins, obtained from different animal sources, and low molecular we...

Antithrombin III deficiency: decreased synthesis of a biochemically normal molecule.

A 29-yr-old white female has suffered from recurrent venous thromboses over the last 12 yr. Plasma antithrombin III (AT-III) levels were 48% of normal by immunoelectrophoresis and 56% by chromogenic assay. Three of four siblings and the father had similar AT-III levels without associated venous thromboses. Heparin-Sepharose chromatography demonstrated normal behavior of the patient's AT-III. He...

The Relative Importance of Thrombin Inhibition and Factor Xa Inhibition to the Antithrombotic Effects of Heparin

The relative importance of antithrombin and anti-factor Xa activities of heparin fractions required to achieve optimal antithrombotic effects is unknown. To study this. we measured the effects of standard heparin. an octasaccharide heparin fraction (anti-factor Xa activity only). and dermatan sulfate (antithrombin activity only) on the prevention of thrombosis and related this to their anticoag...

Automated amidolytic method for determining heparin, a heparinoid, and a low-Mr heparin fragment, based on their anti-Xa activity.

Using the chromogenic substrate S-2222, we have optimized and automated an amidolytic assay for heparin. The assay is based on the detection of anti-Xa activity generated by heparin in plasma. The method is reproducible (intra- and interassay CVs of 2.4 and 3.3%, respectively) and reliable in antithrombin III-deficient plasma. Results of this assay, obtained for plasma samples from patients and...

Structure and activity of a unique heparin-derived hexasaccharide.

A hexasaccharide representing a major sequence in porcine mucosal heparin has been enzymatically prepared from heparin. Its structure was determined by an integrated approach using chemical, enzymatic, and spectroscopic methods. Two-dimensional 1H homonuclear COSY, C-H correlation NMR, and selective irradiation were used to assign many of the NMR resonances. In addition, new techniques includin...
